AbstractThe transport of cargo in axons is essential for the normal functioning, growth and signaling in neurons. Axonal transport is multimodal, involving a combination of active (directed transport mediated by motor proteins) and passive (diffusive, pausing) mechanisms. In this thesis, we study how cargo transport is spatially and temporally modulated in neurons. In the first part of the pre-synopsis talk, I will discuss the slow axonal transport of a small, soluble protein and how the transport is modulated across developmental stages in Drosophila larva. In the second part of the talk, I will discuss how cargo motion properties are tuned to regulate the flow of vesicles in axons. I will show how increased reversals and track-switching events contribute to a higher flow of vesicle cargo and decreased reversals lead to the formation of stationary vesicle clusters that sequester vesicles at locations of static and dynamic obstacles, thus reducing the overall cargo flow. Finally, I will discuss how the cargo is differentially transported in distinct regions in neurons.
